What Glutathione Actually Does, and Why Absorption Is the Whole Game
Your body's master antioxidant, in plain terms
Glutathione is a small molecule assembled from three amino acids: cysteine, glycine, and glutamate. Your cells build it internally in a two-step, energy-dependent process, and research on glutathione synthesis shows the enzyme glutamate-cysteine ligase sets the pace, with cysteine availability acting as the limiting ingredient. Levels also change with age. A study in the American Journal of Clinical Nutrition found that older adults produced glutathione at substantially lower rates than younger controls, and that supplying the missing amino acid building blocks restored production within two weeks. Declining glutathione isn't a fixed feature of getting older, it's a supply problem. People notice it as skin that doesn't bounce back, thinking that feels a step slower, and hangovers that hit harder than they used to.
Why most glutathione never reaches your cells
Here's the part the label doesn't tell you: swallowing glutathione and absorbing it are two different events. Plain glutathione is a fragile tripeptide, and your digestive tract treats it like any other protein fragment, taking it apart before it reaches circulation intact.
The human research reflects that difficulty. A four-week trial of 500 mg of standard oral glutathione twice daily found no significant change in oxidative stress markers or glutathione status compared with placebo. A longer six-month trial at higher intakes did raise body stores of glutathione across blood, red cells, and lymphocytes. What to Avoid When Taking Glutathione
Under-eating protein

This is the most common and least obvious mistake. Glutathione is built from amino acids, and cysteine is the scarce one. When healthy adults ate a diet stripped of sulfur-containing amino acids for ten days, their glutathione synthesis rate fell significantly even though their measured blood concentration looked normal the whole time.
Low protein intake caps how much glutathione your body can build, no matter what you're supplementing. If you eat lightly, skip meals, or eat on a deadline most days, fix that first. Eggs, poultry, fish, dairy, and cruciferous vegetables all supply the sulfur-containing amino acids your cells need.
Skipping the minerals and B vitamins that make it work
Glutathione doesn't act alone. It works through enzymes, and those enzymes need their own nutrients. The glutathione peroxidase family, which neutralizes peroxides using glutathione as fuel, is built around selenium at the catalytic site. Without adequate selenium, the enzyme can't be assembled properly.
Riboflavin matters for the same reason on the recycling side. Glutathione reductase, the enzyme that converts spent glutathione back to its active form, runs on FAD, which your body makes from riboflavin, vitamin B2. Selenium and riboflavin are what let your body reuse glutathione instead of burning through it once and losing it.
Alcohol and acetaminophen on the same day
Both of these spend glutathione directly, which puts them in direct competition with the dose you just took. Acetaminophen metabolism produces a reactive byproduct called NAPQI, and your liver neutralizes it by binding it to glutathione. Every dose draws down the account.
Alcohol works the other end of the same problem. The last thing to avoid is the one you can't feel: a delivery form that doesn't survive the trip. Acetylated glutathione, also called S-acetyl glutathione, is the best-studied of these. Glutathione has an unusually forgiving profile. Your body produces it constantly, and it's present in ordinary foods. The practical limit on glutathione isn't tolerance, it's diminishing return. Past the point your enzymes and cofactors can use it, more isn't doing more.
